Effect of PU.1 on Dectin-1 activation. a HEK-293 T cells were cotransfected with the indicated amount of pRK5-PU.1, 1 μg pGL3-hDectin1, and 10 ng pRL8-SV40 plasmids, and pGL3-basic and pRK5-HA were used as the negative controls. At 24 h post-transfection, the cells were harvested, lysed, and assayed for luciferase activity. The resultant ratios were normalized to the fold change values obtained from HEK-293 T cells cotransfected with pGL3-basic and pRK5-HA. b Cell lysates were analyzed by immunoblotting with rabbit anti-PU.1 antibodies to detect the expression of pRK5-PU.1. β-Tubulin was used to verify equal loading of proteins in each lane. The experiments were repeated at least thrice, and the data from one representative experiment with two technical repeats were presented (*, p < 0.05)
