From: PU.1 is involved in the immune response to Aspergillus fumigatus through upregulating Dectin-1 expression

Expression of Dectin-1 and PU.1 in A. fumigatus conidia-stimulated THP-1 cells. Total RNA were prepared from the stimulated or unstimulated cells at various time points, and real-time PCR was performed to measure the mRNA levels of Dectin-1 (a) and PU.1 (b) at 2, 8, and 12 h after stimulation, respectively. Induction of Dectin-1 and PU.1 expression. Cell lysates were prepared and subjected to SDS-PAGE and Western blot analysis with antibodies against Dectin-1 (c), PU.1 (d), or β-tubulin at the indicated time points. The ratio of Dectin-1 and PU.1 pixel intensity to β-tubulin were analyzed by Image J software (Version 1.39u). The experiments were repeated at least thrice, and the data from one representative experiment with two technical repeats are presented (*, p < 0.05)
