Role of p-38-MAPK and Syk in Mtb -induced ROS production by PMN. (A) PMN were pretreated with DHR and incubated with specific p38 MAPK and Syk inhibitors, SB203580 and piceatannol, respectively (SB and Pic) for 30 min. previous to bacteria challenge (1:2 Mtb:PMN ratio for 90 minutes). Thereafter, the emission of oxidized DHR was evaluated by flow cytometry. Results are expressed as mean ± SE (n = 20); Ra alone vs. all conditions: ***p < 0.001. (B) Percentage of cells that express the activated form of cytoplasmic p38 MAPK and Syk. PMN were incubated with Ra or M strains with neutralizing anti-dectin-1 or corresponding isotype.
